An analysis of the spontaneous mutation rate measurement in filamentous fungi Genet. Mol. Biol.

Mutations related to gene methG1 of Aspergillus nidulans were analyzed, in order to study a mathematical model for the determination of the mutation rate per nucleus per generation, in filamentous fungi. A replica plating technique was used to inoculate, in a single operation, 26 colonies of the strain, into Petri dishes containing culture medium, and the nine central colonies were analyzed for size and number of conidia in each colony. Using this technique, several central colonies were analyzed with regard to the appearance of mutation, and the number and type of reversions were determined for each colony. The frequencies obtained for each reversion were analyzed, in order to verify if their distribution was in accordance with that of Greenwood and Yule....

Genetic structure and demographic history of striped weakfish Cynoscion guatucupa (sciaenidae) from the southwestern atlantic JBAG

In South America, the Pleistocene was characterized by important environmental changes related to glacial cycles, which had significant effects on the evolutionary history of several marine species. To evaluate the pattern of demographic history and the genetic structure of striped weakfish Cynoscion guatucupa, a 401 bp fragment of the mitochondrial Cytochrome b gene was sequenced from 92 individuals from three coastal areas in Argentina and one in Brazil in the southwestern Atlantic. Haplotype diversity was high, whereas nucleotide diversity was low among all sampling sites. The star-like pattern was not phylogeographically structured, in agreement with the AMOVA analysis. Mutation rate of 7 X-STRs of common use in population genetics JBAG

Microsatellite loci analysis is a relevant tool in population genetic studies and also in paternity testing. In this field the X-chromosome STR loci are widely used for female offspring testing but their mutation rates are scarcely known. In this work we performed a mutation rate analysis of seven STR markers located in the X- chromosome specific region, in mother-son pairs and father-mother-daughter trios. Among 1015 allele transfers, we found only one change for a mother-daughter transmission in the DXS9898 locus, consisting of a loss of one single repeat. This finding allowed us to estimate a general mutation rate of μ=9.85 x 10-4 (2.49 x 10-4 - 5.49 x 10-2) which can be applied to population studies. This value is consistent with other reports...
